Mrs Antonio said she was hoping to implement changes through working with the trust after missing out on a position in the last election.
"I hope to see more of a relationship with the community in that they feel the trust is a part of them and representing them," she said.
Lucy Cruickshank said it was an honour to be elected with Mrs Antonio.
"I'm astonished and delighted that the people of Masterton have voted for us women," she said.
She said she would begin her work by learning a lot about the trust and then moving on to "systems and strategies for the future".
"I want to support the community in every way and have prosperity," she said of her visions.
Mrs Antonio also said she had noticed a lack of knowledge during campaigning.
"A lot of comments and feedback I found from campaigning was that many people didn't know what the trust was," she said.
"Being a newcomer with Lucy, I want to make the community aware of what we will be doing,"
Mrs Antonio said she was "pleased with the make-up of the trust".
Mayoral challenger Gary Caffell posted on his Masterton Matters blog that it was a "huge day for ladies".
He then congratulated newly elected mayor Lyn Patterson and Trust Lands Trust member Sandy Ryan, along with Mena Antonio and Lucy Cruickshank.
