At the end of the segment, which also lampoons Trump over the Russia investigation, a message flashes up reminding viewers of the ongoing crisis in Puerto Rico.
The statement reads "Please help for real" and points out that much of Puerto Rico is still without drinking water and electricity.
It's the second time the program has poked fun at the President over his response to the crisis and asked fans to donate to the cause.
People on social media applauded the short clip, with many thanking the creators for drawing attention to the ongoing crisis in the US territory.
Some Twitter users asked how they can help, while a viewer on YouTube predicted that Puerto Rico will reach its goal within a week or two of the episode airing.
The episode also used the Russia investigation to have a further dig at President Trump who has been the butt of many jokes in the series.
The sketch begins with a scene in which Attorney-General Jeff Sessions lures Special Counsel Robert Mueller to a meeting with the President at the Trump Hotel in Washington D.C.
Mueller is heading up the investigation into collusion between Trump's campaign for the White House and the Russians.
After the Special Counsel enters the room The Donald turns on a swivel chair like a Bond villain.
The cartoon president then says: "I do the best bribes, everyone says so. The best! Everyone says so ― the best! Everybody says so!"
The President then pulls a string and a vast wads of cash tumble down from the ceiling.
He continues: "You seem like a very upright guy, how would you like to be a judge? At the Miss Universe pageant!" Trump offers."
But the etching of Mueller refuses and scampers away over the pile of $100 bills.
As soon as the investigator leaves a knock at the door is heard and a housekeeper walks in.
Astonished she asks: "What's this money doing here?"
Trump retorts: "I didn't see any money and neither did you, goodbye I wasn't here,"
The president then runs off through the money.
The housekeeper then smiles and says: "Finally, Puerto Rico catches a break."
She then rips off her housekeeping uniform to reveal the mayoral outfit of San Juan's Mayor Carmen Cruz, who begins to vacuum up the dollars.
Famously the show predicted Trump's unlikely victory all the way back in 2000.
