The British comedian quipped about the struggles of writing the perfect monologue night after night for his hugely popular US show, all while dressed as the chart-topping superstar.
Like Beyonce's Lemonade, LemonJames is split into sections, with James titling his: Trump Joke, Truth Bomb, LemonJames, A New Study, and Hangry.
He moodily tells viewers while sitting in a bathtub that he doesn't want to write any more Donald Trump puns, before referencing the most talked-about line from Beyonce's album.
"I don't want to do another Trump joke. I've exhausted that topic. I've talked about his hair, his tiny baby hands. Doing Trump jokes makes me feel dirty. You know what else makes me feel dirty? This bathtub".
"Does that make you laugh like Jimmy with the good hair?" he teased, referencing Beyonce's call-out to "Becky with the good hair".
While Beyonce fans have been wildly speculating about the identity of 'Becky' and her supposed relationship with the singer's husband Jay- Z, James called out his rival late-night hosts Jimmy Fallon and Jimmy Kimmel.
James is also seen dressed in satin underwear in black and white shots as he runs through the perfect recipe for lemonade, before deciding it's easier to head to the shops.
The final section, titled Hangry, sees James donning a yellow dress just like Beyonce's and a blonde wig as he wields a baseball bat, smashing TVs as Beyonce's song Hold Up plays in the background.
Beyonce's new album has sent her fans wild, mainly over the line "Becky with the good hair", who is said to have had a tryst with her man.
Rachel Roy had originally been the number one candidate for the mystery other woman Beyoncé mentions in the eye-opening track, Sorry.
But now some fans now believe former Roc Nation star Rita Ora could be 'Becky with the good hair'.
Fans flocked to social media to accuse the singer of causing trouble in Beyoncé and Jay Z relationship after a recent Snapchat image she posted was highlighted by eagle-eyed followers.
In a series of Snapchat pictures posted on April 21, just days before Beyoncé released her album, Rita was seen wearing a semi-sheer bra which protected her modesty with strategically placed lemons.
While the bra was in fact one of many fruit motif items from her collaboration with lingerie label, Tenezis, it was Rita's choice of necklace that truly had 'Queen Bey's fans - who call themselves the Beyhive - buzzing with disdain.
The singer wore a gold necklace with the initial 'R', although due to the mirror reflection, some were falsely led some to believe the letter was a 'J'.
